(A) A live ReAsH-stained C2C12 cell transfected with the FL construct has bright puncta for 5 min before the signal is no longer detectable because of "run off" of the labeled nascent polypeptides. The images were a time-lapse series taken every minute starting at 4 min. A schematic representation of the time course of the procedure is provided. (B) In the presence of cycloheximide, an image of a live ReAsH-stained C2C12 cell transfected with the FL construct has bright puncta (translation sites) at 30 min that are still present in the same location after 45 min after removal of ReAsH.
