1. Translation sites do not move.
Time-lapsed video of a ReAsH (red) stained C2C12 cell containing the FL construct. Images were acquired every 5 min for 1 h with a 35 min/s display rate. Notice that the translation sites do not move during the 1-h period, suggesting that they are anchored during the process of translation. See Fig. 4 B.
Movie 2. Newly synthesized β-actin accumulates at cell contacts.
Time-lapsed video of a C2C12 cell containing the FL construct pulse labeled for 1 h with FlAsH and chase labeled for 15 min with ReAsH. The image is a pseudocolored ratio of the red channel to the green channel, with red indicating the areas with high concentrations of newly synthesized β-actin and blue indicating low concentrations of newly synthesized β-actin. Images were acquired every minute for 10 min and displayed at 5 min/s. Notice that β-actin accumulates at the site of cell contact (asterisk) throughout the duration of the experiment. See Fig. 7 F.
